Function
- polycomb group (PcG) protein
- component of the PRC2/EED- EZH2 complex, which methylates Lys-9 & Lys-27 of histone H3, leading to transcriptional repression of the affected target gene
- the PRC2/EED-EZH2 complex may also serve as a recruiting platform for DNA methyltransferases, thereby linking two epigenetic repression systems
- genes repressed by the include HOXC8, HOXA9, MYT1 & CDKN2A
- interacts with MLL (putative)
- component of the PRC2/EED-EZH2 complex
- interacts with HDAC, HDAC2, histone H1 & YY1
- may interact with ITGA4, ITGAE & ITGB7
- may interact with the MA protein of HIV-1
Structure
- belongs to the WD repeat ESC family
- contains 7 WD repeats
Compartment
- nucleus
- transiently colocalizes withXIST at inactive X chromosomes
Alternative splicing
- named isoforms=3
- additional isoforms may be produced by alternative initiation, both from non-canonical start codons upstream of the initiator Met displayed & from other canonical start codons downstream of that displayed; the precise sites of translation initiation have not been unambiguously identified
Expression
- expressed in brain, colon, heart, kidney, liver, lung, muscle, ovary, peripheral blood leukocytes, pancreas, placenta, prostate, spleen, small intestine, testis, thymus & uterus.
- expression peaks at the G1/S phase boundary
- expression is induced by E2F1, E2F2 & E2F3
Pathology
- appears to be overexpressed in breast & colon cancer
