Function
- transcriptional activator involved in T-cell lymphocyte differentiation
- necessary for the survival of CD4(+) CD8(+) immature thymocytes deficiency blocks transition from CD8+ to CD8+/CD4+ T-cells
- isoforms lacking the N-terminal CTNNB1 binding domain cannot fulfill this role
- binds to the T-lymphocyte-specific enhancer element (5'-WWCAAAG-3') found in the promoter of the CD3E gene
- regulates activity of the T-cell Receptor-alpha gene enhancer
- may also act as feedback transcriptional repressor of CTNNB1 & TCF7L2 target genes
- TLE1, TLE2, TLE3 & TLE4 repress transactivation mediated by TCF7 & CTNNB1
- binds the armadillo repeat of CTNNB1 & forms a stable complex
- interacts with AES, TLE1, TLE2, TLE3 & TLE4
Structure
- belongs to the TCF/LEF family
- contains 1 HMG box DNA-binding domain
- very limited homology to the ETS domain exists within the HMG box

- additional isoforms seem to exist Alternative promoter:
- 2 series of isoforms, L & S, are produced by use of alternative promoter usage
Expression
- expression is complex & overlapping during embryogenesis, but is restricted largely to lymphocytes postnatally
- expression largely restricted to the thymus
- expressed predominantly in T-cells
- induced following T-cell activation
- also detected in proliferating intestinal epithelial cells & in basal epithelial cells of mammary gland epithelium
- induced by TCF7L2 & CTNNB1
