Function
- may be involved in regulating immune reactivity
- may play a role in modulating dendritic cell differentiation &/or maturation
- may be involved via its ITIM motif in the inhibition of B-cell-receptor-mediated Ca+2 mobilization & protein tyrosine phosphorylation
- may interact with PTPN6 via its ITIM motif
Structure
- contains 1 ITIM motif
- the phosphorylated ITIM motif can bind the SH2 domain of several SH2 domain-containing phosphatases
- contains 1 C-type lectin domain
Compartment
- membrane; single-pass type 2 membrane protein (probable)
Alternative splicing
named isoforms=4
Expression
- expressed in dendritic cells, myeloid cells, B-cells & HL-60 cells (at protein level)
- TNF alpha, IL-1 alpha, & LPS, down-regulate expression at the surface of neutrophils (at protein level)
- expressed preferentially in hematopoietic tissues
- expressed in peripheral blood leukocytes, neutrophils
- moderate expression in spleen, lymph node, & bone marrow
- very low expression in thymus
- expressed in Ag-presenting cells (dentritic cells, monocytes, macrophages & B-cells)
- expressed on granulocytes
- expression decreased in dentritic cells by signals inducing maturation (e.g. CD40 ligand, LPS, & TNF alpha)
- isoform 2 mRNA expression is up-regulated by agonists of neutrophils GM-CSF, IL-3, IL-4 & IL-13
