Function
- mediates homophilic cell-cell adhesion (Ca+2-independent)
- also mediates heterophilic cell-cell adhesion with CADM3 & PVRL3 (Ca+2-independent)
- acts as tumor suppressor in non-small-cell lung cancer cells
- interaction with CRTAM promotes
- NK cell cytotoxicity & IFN-gamma secretion by CD8+ cells in vitro
- NK cell-mediated rejection of tumors expressing CADM3 in vivo
- may contribute to the less invasive phenotypes of lepidic growth tumor cells
- in mast cells, may mediate attachment to & promote communication with nerves
- CADM1, together with MITF, is essential for development & survival of mast cells in vivo
- synaptic cell adhesion molecule that plays a role in formation of dendritic spines & drives synapse assembly
- role in neuronal migration, axon growth, pathfinding, & fasciculation on the axons of differentiating neurons
- glycosylation at Asn-67 & Asn-101 promotes adhesive binding & synapse induction
- diverse roles in spermatogenesis including
- adhesion of spermatocytes & spermatids to Sertoli cells
- normal differentiation into mature spermatozoa
- interacts with CRTAM
- interacts with EPB41L3/DAL1 via C-terminus
- interaction with EPB41L3/DAL1 may act to anchor CADM1 to actin cytoskeleton
- interacts via its C-terminus with the PDZ domain of MPP3 & the PDZ domain of MPP6.
Structure
- homodimer
- cytoplasmic domain appears to play a critical role in proapoptosis & tumor suppressor activity in NSCLC
- belongs to the nectin family
- contains 2 Ig-like C2-type domains (immunoglobulin-like)
- contains 1 Ig-like V-type domains (immunoglobulin-like)
Compartment
- cell membrane
- cell junction, synapse
- associates with perinuclear & plasma membranes in vivo
- localized to the basolateral plasma membrane of epithelial cells in gall bladder
Alternative splicing
named isoforms=5
Pathology
- absent or down-regulated in many advanced cases of NSCLC as well as in many other human cancers, due to gene silencing by promoter methylation
- may contribute to less invasive phenotypes of lepidic growth tumor cells
More general terms
- immunoglobulin superfamily protein
- glycoprotein
- membrane protein
- anti-oncoprotein (tumor suppressor protein)
- human longevity protein
