- the VEGF-kinase ligand/receptor signaling system plays a role in vascular development & regulation of vascular permeability
- interacts with SHB upon VEGF activation
- Flk1 & Flt1 bind VEGF with high affinity & are required for embryonic & definitive hematopoiesis

- expressed in developing blood islands of primitive streak stage embryos
- expressed in posterior splanchnopleure & fetal liver hematopoietic precursors
Pathology
- interacts with HIV-1 Tat
- in case of HIV-1 infection, the interaction with extracellular viral Tat protein seems to enhance angiogenesis in Kaposi's sarcoma lesions
Pharmacology
- ramucirumab used for treatment of advanced gastric cancer,
- gastroesophageal junction adenocarcinoma & metastatic colorectal cancer inhibits VEGFR2 via its ligand-binding site
