Function
- role in terminally differentiating adult nervous system during postnatal development
- may stabilize interactions between hyaluronan & brain proteoglycans
- brevican is integral to the formation of the extracellular matrix's neuroprotective perineuronal nets[2]
- interacts with TNR
- biomarker of brain aging (negative correlation)
Structure
- contains mostly chondroitin sulfate
- isoform 2 is predicted to have its GPI-anchor on Asn-649
- belongs to the aggrecan/versican proteoglycan family.
- contains 1 C-type lectin domain
- contains 1 EGF-like domain
- contains 1 Ig-like V-type domain (immunoglobulin-like)
- contains 2 Link domains
- contains 1 Sushi domain (CCP/SCR)
Compartment
- isoform 1: extracellular matrix
- isoform 2: membrane GPI-anchor
Alternative splicing
named isoforms=2
Expression
- isoform 1:
- highly expressed from birth through 8 years of age
- down-regulated by 20 years of age
- low levels in normal adult cortex
- isoform 2
- expressed at uniformly low levels throughout development
Pathology
- lower MRI white matter hyperintensities associated with lower serum brevican
- brevican is reduced in temporal cortex soluble fractions & frontal cortex synaptosomal fractions in Alzheimer's disease[3]
- CSF levels of brevican positively correlates with age, total tau, p-Tau, neurofilament-L & A-beta40[3]
