Structure
- composed of alternating sulfated N-acetyl-galactosamine & glucuronate residues (1,3 beta linkage)
Biochemistry
- chondroitin sulfate constitutes the predominant proteoglycan present in cartilage & is distributed on the surfaces of many cells & extracellular matrix
- carbohydrate sulfotransferase 3 (CHST3) sulfates chondroitin to form chondroitin-6-sulfate
- major excreted mucopolysaccharide in normal individuals is chondroitin-6-sulfate
- chondroitin-4-sulfate is found is association with inter-alpha-trypsin inhibitor
- carbohydrate sulfotransferases (CHST11, CHST12, CHST13) sulfate chondroitin to form chondroitin-4-sulfate
Pharmacology
- used for treatment of osteoarthritis
- often combined with glucosamine
- Refs[5][6] suggest combined with glucosamine no better than glucosamine alone
- Ref[7][8] suggests chrondroitin useless
- no benefit of glucosamine or chondroitin for attenuating joint space narrowing[9]
- may delay progression of knee osteoarthritis; however, pain benefits not clear[10]
- neither glucosamine, chondroitin or combination of the two beneficial in osteoarthritis of the knee[12]
- pharmaceutical-grade chondroitin sulfate 800 mg/day as effective as celecoxib for osteoarthritis of the knee[13]
- may benefit hand osteoarthritis[9]
