Function
- calmodulin mediates the control of a large number of enzymes, ion channels, aquaporins & other proteins through Ca+2-binding
- among the enzymes to be stimulated by the calmodulin-Ca+2 complex are a number of protein kinases & phosphatases
- together with CCP110 & centrin, is involved in a genetic pathway that regulates the centrosome cycle & progression through cytokinesis
- interacts with MYO1C, MYO5A & RRAD
- interacts with MYO10 (putative)
- interacts with CEP97, CCP110, TTN/titin & SRY
- interacts with USP6, CDK5RAP2, SCN5A (putative)
- interacts with RYR1, FCHO1
- interacts with MIP, promotea MIP water channel closure (putative)
- interacts with ORAI1, IQCF1, SYT7 (putative)
- interacts with CEACAM1, RYR2, PCP4
- interacts with the heterotetrameric KCNQ2 & KCNQ3 channel
- ubiquitination results in a strongly decreased activity
- phosphorylation results in a decreased activity
Structure
- belongs to the calmodulin family
- 4 functional Ca+2-binding sites
Compartment
- cytoplasm, cytoskeleton, spindle, spindle pole
- distributed throughout the cell during interphase
- during mitosis becomes localized to the spindle poles & the spindle microtubules
Pathology
- catecholaminergic polymorphic ventricular tachycardia is a rare condition caused by mutation in CALM3
