Function
- adapter protein
- negatively regulates T-cell receptor (TCR) signaling
- inhibits T-cell antigen-receptor induced activation of NFAT
- role in negative regulation of positive selection & mitosis of T-cells
- links signaling proteins such as ZAP70 with CBL, leading to CBL dependent degradation of signaling proteins (putative)
- interacts with EPHA2, VAV1, LCP2, PDGFRB (putative)
- homodimerization & interaction with phosphorylated CBL occurs via its C-terminal domain
- interacts with phosphorylated proteins ZAP70, CD3Z, SYK & LAT via its SH2 domain
- indirectory induced by all-trans retinoic acid
- C-terminal domain is essential for
- homodimerization (via charged region of domain)
- interaction with CBL (via hydrophobic region of domain)
Structure
- homodimer
- phosphorylated
- contains 1 SH2 domain
- contains 1 SH3 domain
Compartment
Expression
- expressed in lung & fetal brain > heart, adult brain, placenta, liver, skeletal muscle, kidney, pancreas
