Function
- tumor suppressor (putative)
- death receptor-dependent apoptosis
- when associated with MOAP1, promotes BAX translocation to mitochondrial membranes in response to TNF & TNFSF10
- inhibits proliferation
- negatively regulates cell cycle progression G1/S-phase transition - regulates cyclin D1 protein
- inhibits activation of STK3 & STK4
- scaffold protein (putative)
- interacts with XPAl, CDC20 (during prometaphase), STK3, STK4, NFRSF1A, TNFRSF10A, RSSF5, HRAS1, RSSF1, XPA
Compartment
- isoform A localizes to microtubules during interphase, centrosomes associated with microtubules during prophase, spindle fibers & spindle poles at metaphase & anaphase, midzone during early telophase, midbody in late telophase in cells
- isoform C is predominantly nuclear
Alternative splicing
7 alternate splice forms, A-G
Expression
- isoform A & isoform C are ubiquitously expressed
- isoform B is expressed in hematopoietic cells
Pathology
- loss of RASSF1A seen in small cell lung cancer & some other cancer cell lines which correlate with hypermethylation of the CpG island promoter sequence of RASSF1A
