Function
- isoform 1 acts as an E3 ubiquitin ligase, which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, & then transfers it to substrates promoting their degradation by the proteasome
- promotes degradation of TLR4 & TLR9
- isoform 3/ZIN inhibits TNF & IL-1 mediated activation of NF-kappa-B
- promotes TNF & RIP mediated apoptosis
- protein modification; protein ubiquitination
- auto-ubiquitinated
- interacts with UBE2L3 & to some extent with UBE2L6
- interacts with TLR3, TLR4, TLR5 & TLR9
- isoform 3/ZIN binds RIPK1 & HIV VIF
Structure
- the RING-type Zn+2 finger domain mediates binding to an E2 ubiquitin-conjugating enzyme (putative)
- contains 1 IBR-type Zn+2 finger
- contains 2 RING-type Zn+2 fingers
Compartment
Alternative splicing
Expression
- ubiquitous
- highest levels of expression in testis & peripheral blood leukocytes
