Function
- facilitates polymerization of neurofilaments from individual subunits NEFH & NEFL
- required for organization of the cellular microtubule array & microtubule anchoring at the centrosome
- regulates microtubule organization at least in part by targeting the microtubule severing protein KATNA1 to the centrosome (putative)
- positively regulates activity of minus-end directed microtubule motor protein dynein
- enhances dynein-mediated microtubule sliding by targeting dynein to microtubule plus ends (putative)
- required for several dynein- & microtubule-dependent processes, including maintenance of Golgi, centripetal motion of secretory vesicles & coupling of the nucleus & centrosome
- required during brain development for migration of newly formed neurons from ventricular/subventricular zone toward the cortical plate
- dispensible for mitosis in cortical neuronal progenitors, which instead requires NDE1
- interacts with YWHAE
- interacts directly with NEFL & indirectly with NEFH
- interacts with microtubules
- self-associate
- interacts with DISC1, dynein, dynactin, tubulin gamma, KATNA1, KATNB1, PAFAH1B1, PCM1 & PCNT
- phosphorylated in mitosis
- can be phosphorylated by CDC2, CDK5 & MAPK1
- phosphorylation by CDK5 promotes interaction with KATNA1 & YWHAE
Structure
- belongs to the nudE family
Compartment
- localizes to the cell body of motor neurons
- colocalizes with assembled neurofilaments within axonal processes
- localizes to microtubules of the manchette in elongated spermatids
- cytoplasm
- localizes to interphase centrosome & mitotic spindle
Alternative splicing
named isoforms=2
Expression
- expressed in brain, heart, kidney, liver, lung, pancreas, placenta & skeletal muscle
- expression peaks in mitosis
