Introduction
Founding member of the cyclin-dependent kinase family.
Function
- CDK1 is a component of MPF (Maturation Promoting Factor) & of CTD kinase
- role in the control of the eukaryotic cell cycle
- required in higher cells for entry into S-phase & mitosis
- p34 is a component of the kinase complex that phosphorylates the repetitive C-terminus of RNA polymerase 2
- forms a stable but non-covalent complex with a regulatory subunit & with a cyclin
- interacts with DLG7
- isoform 2 is unable to complex with cyclin B1 & also fails to bind to the CDK inhibitor p21
- interacts with catalytically active CCNB1 & RALBP1 during mitosis to form an endocytotic complex during interphase
- phosphorylation at Thr-14 or Tyr-15 inactivates the enzyme
- phosphorylation at Thr-161 activates it
- component of MPF (Maturation Promoting Factor)
- component of CTD kinase.
Structure
- belongs to the protein kinase superfamily, CMGC Ser/Thr protein kinase family, CDC2/CDKX subfamily
- contains 1 protein kinase domain
Compartment
Alternative splicing
named isoforms=2
Pathology
found in breast cancer tissues
Comparative biology
- Yeast homologs are cdc2 (S. cerevisiae), cdc28 (S. pombe), cdc2-kinase
- Thr-14 phosphorylation site is not present in yeast homologs
