Function
- responds to activation by environmental stress & pro-inflammatory cytokines by phosphorylating downstream targets
- role in regulation of protein translation by phosphorylating & inactivating EEF2K
- activated by phosphorylation on Thr & Tyr by either of two dual specificity kinases, MAP2K3 & MAP2K6
- dually phosphorylated on Thr-180 & Tyr-182, which activates the enzyme
Structure
- TXY motif contains the Thr & Tyr whose phosphorylation activates the MAP kinases
- belongs to the protein kinase superfamily, CMGC Ser/Thr protein kinase family, MAP kinase subfamily
- contains 1 protein kinase domain
Expression
- expressed in testes, pancreas, small intestine, lung & kidney
- abundant in macrophages
- also present in neutrophils, CD4+ T-cells, & endothelial cells
