Function
- seeems to be involved in transcriptional regulation
- DAXX seems to regulate transcription in PML nuclear bodies together with transcription factor PML & may influence TNFRSF6-dependent apoptosis
- role in checkpoint responses upon DNA damage[3]
- heterochromatin remodelling in G2 phase[4]
Structure
- diameters between 0.2 & 1 mm
- number & size change during the cell cycle
- electron microscopic analysis show that the nuclear body is shaped like a doughnut
- components of the nuclear body include:
- transcription factor PML
- sumoylation is necessary for PML nuclear body formation
- nuclear body protein SP140
- RB protein
- protein p53
- CBP
- DNA helicase BLM22
- eIF-4
- INT-6
- Daxx
- SUMO-1
- HAUSP
- Sp100
- transcription factor PML
Expression
Notes
- discrete nuclear speckles containing transcription factor PML along with other proteins, which were originally identified as autoantigens in primary biliary cirrhosis patients
