Function
- homophilic clustering adhesion molecule
- enhances interferon gamma secretion in lymphocytes
- recruits SH2 domain-containing proteins SH2D1A/SAP
- increases proliferative responses of activated T-cells
- induces platelet stimulation (SH2D1A/SAP-dependent)
- marker for hematopoietic progenitor cells
- interacts with SH2 domain-containing proteins SH2D1A/SAP & SH2D1B/hEAT-2
- interaction with tyrosine-protein phosphatases PTPN6 & PTPN11 via its phosphorylated cytoplasmic domain, blocked by SH2D1A
- ITSM motif may bind SH2D1A
- phosphorylated by tyrosine-protein kinase LCK on Tyr following ligation induced by agonist monoclonal antibody
- association with SH2D1A/SAP is dependent on Tyr phosphorylation of its cytoplasmic domain
- phosphorylated on Tyr-296 & Tyr-316 following platelet aggregation
Structure
- homooligomer, via its extracellular domain
- N-glycosylated
- contains 1 Ig-like C2-type domain (immunoglobulin-like)
Compartment
Alternative splicing
named isoforms=7
Expression
- predominantly expressed in hematopoietic tissues, including lymph nodes, spleen, peripheral leukocytes.
- expressed in macrophages, B-cells, monocytes, platelets, thymocytes, T-cells, dentritic cells
- highly expressed in memory T-cells
- expression is slightly increased in naive B-cells after 1st division
- in contrast, expression on memory B-cells decreased with each successive division
