Introduction
Docking proteins interact with receptor tyrosine kinases & mediate particular biological responses.
Function
- RET-mediated neurite outgrowth
- activation of the MAP kinase pathway
- interacts with phosphorylated RET
- link with downstream effectors of RET in neuronal differentiation
- does NOT interact with RasGAP
- phosphorylated on Tyr in response to insulin, IGF1 & GDNF
Structure
- PTB domain mediates receptor interaction (putative)
- belongs to the DOK family, type B subfamily
- contains 1 IRS-type PTB domain
- contains 1 PH domain
Alternative splicing
named isoforms=2
Expression
- highest expression in skeletal muscle
- lower in brain, heart & kidney
- detected in activated peripheral blood T lymphocytes
