Introduction

The phosphotyrosine-binding domain PTB is a domain of about 200 amino acids in length which bears little resemblance to SH2 domains. The PTB domain appears to recognize motifs with the consensus Asn-Pro-C-pTyr. Thus PTB contrasts with the SH2-domain, in requiring residues N-terminal to the pTyr. PTB appears important for the interactions of the Shc protein and IRS-1 with activated receptors.
