Function
- cell adhesion & spreading factor in plasma & tissues.
- interaction with glycosaminoglycans & proteoglycans
- recognized by some integrins
- inhibitor of the membrane-damaging effect of the terminal
- interacts with SERPINE1/PAI1 & insulin cytolytic complement pathway
- phosphorylation on Thr-69 & Thr-76 favors cell adhesion & spreadings
- precursor for somatomedin B
Structure
- exists in two forms: a single chain 75 kD form (V75) & a clipped form composed of two chains (65 kD & 10 kD) (V65+V10) which are held together by a disulfide bond
- sulfated on 2 Tyr, N- & O-glycosylated (putative)
- SMB domain may have disulfide bond heterogeneity
- SMB domain mediates interaction with SERPINE1/PAI1
- heparin-binding domain mediates interaction with insulin
- contains 4 hemopexin-like domains
- contains 1 SMB (somatomedin-B) domain
