Function
- receptor for netrin
- involved in axon guidance
- mediates axon repulsion of neuronal growth cones in developing nervous system
- association with DCC (cytoplasmic domain) may trigger signaling for axonal repulsion
- corticospinal tract axon guidance, independent of DCC
- apoptosis induction when NOT associated with netrin
- phosphorylated on different cytoplasmic Tyr
- phosphorylation of Tyr-568 leads to an interaction with PTPN11 phosphatase (netrin-dependent)
- proteolytic cleavage by caspases during apoptosis does NOT occur associated with netrin ligand
- cleavage by caspases is required to induce apoptosis
Structure
- belongs to the unc-5 family
- contains 1 death domain
- contains 1 Ig-like (immunoglobulin-like) domain
