Introduction
Tradename: Pro-Banthine.
Indications
- adjunctive therapy for peptic ulcer disease
- irritable bowel disease
- used in conjunction with H2 receptor antagonists in the treatment of Zollinger-Ellison syndrome
- urinary incontinence[4]
Dosage
Tabs: 7.5 & 15 mg.
Pharmacokinetics
- quaternary ammonium drug with variable GI absorption, depending upon presence of food
- distribution is unknown, but does NOT readily penetrate CNS
- elimination 1/2life is 9 hours
elimination via liver
elimination via kidney
1/2life = 9 hours
Adverse effects
- common (> 10%)
- constipation
- decreased sweating,
- dry mouth, skin, nose or throat
- less common (1-10%)
- difficulty swallowing
- uncommon (< 1%)
- cholinergic effects:
Mechanism of action
- not fully understood
- may act as nonselective smooth muscle relaxant
- little or no muscarinic activity, except at very high doses
