- ERCC1/XPF is a structure-specific nuclease; it cleaves Y-type structures at the junction of DNA double-stranded & single-stranded regions
- ERCC1/XPF cuts on the strand that leads off from the junction in the 5'-3' direction
- ERCC1/XPF probably works in coordination with the XP G-correcting protein in excision repair, the two nucleases cleaving the same DNA strand on opposite sides of DNA damage
