Function
- binds VLDL & transports it into cells by endocytosis
- receptor-ligand complexes cluster into clathrin-coated pits prior to endocytosis
- binding to Reelin induces Tyr phosphorylation of Dab1 & modulation of Tau phosphorylation (putative)
- binds to the extracellular matrix protein Reelin
- interacts with VLDLR
- interacts with SNX17 (putative)
- interacts with DAB1
- receptor for the minor-group human rhinoviruses (HRVs)
- binds protein VP1 through the 2nd & 3rd LDL-receptor class A domains
- VLDL receptor binds apolipoprotein E
Structure
- contains 3 EGF-like domains
- contains 8 LDL-receptor class A domains
- contains 6 LDL-receptor class B repeats
Compartment
- membrane
- clathrin-coated pit
Alternative splicing
named isoforms=2
Expression
- abundant in heart & skeletal muscle
- also expressed in ovary & kidney
- tissue distribution of the VLDL receptor is very different from the LDL receptor
- mRNA for the VLDL receptor is abundant in cells with active fatty acid metabolism, including heart, muscle & adipose tissue
- very little is expressed in liver.
Pathology
- deletions involving VLDLR may be the cause of VLDLR-associated cerebellar hypoplasia
