Function
- component of nuclear complexes:
- CCR4-NOT core complex
- chromatin assembly factor 1 (CAF-1 complex)
- CHAF1A binds directly to CHAF1B within the complex
- CHAF1A binds to histones H3 & H4
- may play a role in heterochromatin maintenance in proliferating cells by bringing newly synthesized cbx proteins to heterochromatic DNA replication foci
- CHAF1A binds PCNA, CBX1, MBD1, CBX5
- during DNA replication, CHAF1A forms a S phase-specific complex that facilitates DNA methylation & histone H3 'Lys-9' methylation during replication-coupled chromatin assembly
Structure
- homodimer
- contains one Pro-Xaa-Val-Xaa-Leu (PxVxL) motif required for interaction with chromoshadow domains
- belongs to the CHAF1A family
Compartment
