Function
- component of vault ribonucleoprotein complex
- down-regulates INFG-mediated STAT1 signaling & subsequent activation of JAK
- down-regulates SRC activity & signaling through MAP kinases
- phosphorylated on Tyr after EGF stimulation
- dephosphorylated by PTPN11 - interacts with APEX1
- may interact with ZNF540
Structure
- MVP 3 mediates interaction with PTEN
- MVP 4 mediates interaction with PARP4
- contains 9 MVP (vault) repeats
Compartment
- cytoplasmic, nucleus
- 5% localizes to the nuclear pore complex
- translocates from the nucleus to the cytoplasm upon EGF treatment
Expression
- expressed in most normal tissues
- higher expression observed in epithelial cells
- up-regulated by IFNG/IFN-gamma
Pathology
- higher expression in cells chronically exposed to xenobiotics, such as bronchial epithelium & intestinal epithelial cells
- overexpressed in many multidrug-resistant cancer cells
