Function

• most abundant & widely expressed PKA regulatory subunit
• pseudophosphorylation site binds to the substrate-binding region of the catalytic chain, thus inhibiting activity
• activation by cAMP produces two active cAMP protein kinase monomers & a PKA1 regulatory chain dimer that binds 4 cAMP
• interacts with RFC2, AKAP4
• may be involved in cell survival
• may act as a tumor-suppressor gene in Carney complex 1 sporadic adrenal & thyroid tumors

Pathology

• defects in PRKAR1A associated with Carney complex 1
• defects in PRKAR1A associated with cardiac myxoma
