Function
- most abundant & widely expressed PKA regulatory subunit
- pseudophosphorylation site binds to the substrate-binding region of the catalytic chain, thus inhibiting activity
- activation by cAMP produces two active cAMP protein kinase monomers & a PKA1 regulatory chain dimer that binds 4 cAMP
- interacts with RFC2, AKAP4
- may be involved in cell survival
- may act as a tumor-suppressor gene in Carney complex 1 sporadic adrenal & thyroid tumors
Pathology
- defects in PRKAR1A associated with Carney complex 1
- defects in PRKAR1A associated with cardiac myxoma
