• role in the TLR & IL-1 signaling pathways via interaction with the complex containing IRAK kinases & TRAF6
• mediates Lys-63-linked polyubiquitination of IRAK1
• can activate the MAP kinase pathway leading to activation of ELK1
• not required for NF-kappa-B activation
• protein modification; protein ubiquitination
• phosphorylated by IRAK1 & IRAK4 enhancing its E3 ligase activity
• interacts with TRAF6, IRAK1, IRAK4 & MAP3K7

• the atypical FHA domain contains a 'wing' insert & mediates binding to Thr-phosphorylated IRAK1
• belongs to the pellino family
