Function
- E3 ubiquitin protein ligase activity
- regulates levels of CASP8 & CASP10 by targeting them for proteasomal degradation
- protects cells against apoptosis induced by TNF.
- binds phosphatidylinositol-5-phosphate & phosphatidylinositol-3-phosphate.
- interacts with DNAJA3 (putative), CASP8, CASP10
- auto-ubiquitinated (in vitro)
- proteolytically cleaved by caspases after induction of apoptosis by TNF
Structure
- contains 1 FYVE-type Zn+2 finger
- contains 1 RING-type Zn+2 finger
- contains 2 SAP domains
Compartment
- nucleus, nucleoplasm, nuclear speckle
- intracellular membrane, peripheral membrane
Alternative splicing
named isoforms=2
Expression
- ubiquitous
- expressed in heart, brain, liver, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis, ovary, colon, leukocytes
Notes
- originally isolated as potential interaction partner of mitochondrial protein DNAJA3, but the 2 proteins are NOT in the same cellular compartments
