Function
- can insert into membranes & form poorly selective ion channels that may also transport chloride ions
- may play role in regulation of transepithelial ion absorption & secretion
- required for normal formation of stereocilia in the inner ear & normal development of the organ of corti (putative)
- required for the development &/or maintenance of proper glomerular endothelial cell & podocyte architecture
- component of a multimeric complex consisting of several cytoskeletal proteins, including actin, ezrin, alpha-actinin, gelsolin, & IQGAP1
- interacts with AKAP9
Structure
- members of this family may change from a globular, soluble state to a state where the N-terminal domain is inserted into the membrane & functions as chloride channel
- a conformation change of the N-terminal domain is thought to expose hydrophobic surfaces that trigger membrane insertion (putative)
- belongs to the chloride channel CLIC family
- contains 1 GST C-terminal domain
Compartment
- isoform 1:
- cytoplasm, cell cortex, cytoskeleton, membrane
- single-pass membrane protein
- associates with the cortical actin cytoskeleton
- exists both as soluble cytoplasmic protein & as membrane protein
- isoform 2
- Golgi, cytoplasm, cytoskeleton, centrosome, membrane
- single-pass membrane protein
- colocalized with AKAP9 at the Golgi & the centrosome (in part)
- exists both as soluble cytoplasmic protein & as membrane protein
- colocalized with podocalyxin in renal glomeruli
Alternative splicing
named isoforms=2
Expression
- isoform 1: expressed in renal glomeruli endothelial cells & podocytes (at protein level)
