Function
- receptor for ADP
- coupled to Gi-proteins
- may play a role in hematopoiesis & the immune system
Structure
- belongs to the G-protein coupled receptor 1 family
Compartment
- cell membrane; multi-pass membrane protein
Alternative splicing
named isoforms=2
Expression
- strong expression in spleen & adult brain
- lower expression in placenta, lung, liver, spinal cord, thymus, small intestine, uterus, stomach, testis, fetal brain, & adrenal
- not detected in pancreas, heart, kidney, skeletal muscle, ovary or fetal aorta
- detected in lymph node & bone marrow
- weakly detected in peripheral blood mononuclear cells
- but not detected in polymorphonuclear leukocytes
- detected in all brain regions examined
Notes
- stimulation by ADP in stably transfected CHO cells results in inhibition of adenylyl cyclase & phosphorylation of MAP kinases MAPK3 & MAPK1 (pertussis toxin-sensitive)
- inhibition of adenylyl cyclase & phosphorylation of MAP kinases are signal transduction mechanisms that involve Gi proteins
