• role as a marker & a regulator of neuronal differentiation
• up-regulated byvariety of neurogenic signals, including retinoic acid, EGF & NGF
• induces apoptosis, growth arrest & expression of cyclin- dependent kinase inhibitor CDKN1B
• role as tumor repressor
• inhibits cell growth & tumorigenicity of medulloblastoma
• antagonist of Hedgehog pathway on cell proliferation & differentiation by affecting nuclear transfer of transcription factor GLI1
  • maintains cerebellar granule cells in undifferentiated state
• activates the caspase cascade

Structure

• contains 1 BTB domain (POZ domain)
  • BTB domain is required for growth-suppressing properties

Expression

• higher expression in cerebellum than in whole brain

Pathology

• underexpression in medulloblastoma.
• haploinsufficiency of KCTD11 may be a cause of medulloblastoma
