Function
- role as a marker & a regulator of neuronal differentiation
- up-regulated byvariety of neurogenic signals, including retinoic acid, EGF & NGF
- induces apoptosis, growth arrest & expression of cyclin- dependent kinase inhibitor CDKN1B
- role as tumor repressor
- inhibits cell growth & tumorigenicity of medulloblastoma
- antagonist of Hedgehog pathway on cell proliferation & differentiation by affecting nuclear transfer of transcription factor GLI1
- maintains cerebellar granule cells in undifferentiated state
- activates the caspase cascade
Structure
- contains 1 BTB domain (POZ domain)
- BTB domain is required for growth-suppressing properties
Expression
- higher expression in cerebellum than in whole brain
Pathology
- underexpression in medulloblastoma.
- haploinsufficiency of KCTD11 may be a cause of medulloblastoma
