Function
- MEK5 & ERK5 interact specifically with one another
- activated by MEK5
- does not interact witwith MEK1/ERK1 or MEK2/ERK2 pathways
- may interact with actin to target to specific cell location
- activated by Tyr & Thr phosphorylation (putative)
- dually phosphorylated on Thr-218 & Tyr-220, which activates the enzyme (putative)
- autophosphorylated in vitro on Thr & Tyr when the C-terminal part of the kinase, which could have a regulatory role, is absent
Structure
- 2nd proline-rich region may interact with actin targeting the kinase to a specific location in the cell
- TXY motif contains the Thr & Tyr whose phosphorylation activates the MAP kinases
- belongs to the protein kinase superfamily, CMGC Ser/Thr protein kinase family, MAP kinase subfamily
- contains 1 protein kinase domain
Expression
- expressed in many adult tissues
- abundant in heart, placenta, lung, kidney & skeletal muscle
- not detectable in liver
