• activated by phosphorylation
• phosphorylation on Ser/Thr by MAP kinase kinase kinases (RAF or MEKK1) regulates positively the kinase activity
• acetylation by Yersinia yopJ prevents phosphorylation & activation, thus blocking MAPK signaling pathway
