Function
- adapter protein between membrane proteins & actin cytoskeleton
- role in receptor clustering & cytoskeletal polarity in junction between T-cell & antigen-presenting cell
- may anchor podocyte slit diaphragm to the actin cytoskeleton in renal glomerulus
- interacts through its second SH3 domain with phosphorylated CBL C-terminus
- interacts with BCAR1/p130Cas SH3 domain
- interacts with F-actin, CD2, PKD2, NPHS1 & NPHS2
- interacts with FAM125A
- phosphorylated on tyrosine residues, probably by c-Abl, Fyn & c-Src
Structure
- homodimer, mediated by coiled coil domain
- contains 3 SH3 domains
- Pro-rich domain may mediate binding to SH3 domains
Compartment
- cytoplasm
- colocalizes with F-actin & BCAR1/p130Cas in membrane ruffles
- located at podocyte slit diaphragm between podocyte foot processes
Expression
widely expressed in fetal and adult tissues
Pathology
- defects in CD2AP are the cause of susceptibility to focal segmental glomerulosclerosis 3
