zinc finger FYVE domain-containing protein 9; mothers against decapentaplegic homolog-interacting protein; Madh-interacting protein; smad anchor for receptor activation; receptor activation anchor; hSARA; Novel serine protease; NSP (ZFYVE9, MADHIP, SARA, SMADIP)

Function
- SARA binds with SMAD2 & SMAD3 & TGF-beta receptor, interacting with MH2 domain of SMAD2 & SMAD3
- phosphorylation of SMAD2 by TGF-beta receptor induces dissociation from SARA & formation of SMAD2/SMAD4 complexes & nuclear translocation
- role in recruiting unphosphorylated forms of SMAD2/SMAD3 to the TGF-beta receptor by controlling their subcellular localization & by interacting & colocalizing with the TGF-beta receptor
Structure
- FYVE domain mediates binding to phospholipids & targets SMADs to endosomes
Compartment
Alternative splicing
- named isoforms=3
- at least one isoform may be produced at very low levels due to a premature stop codon in the mRNA, leading to nonsense-mediated mRNA decay
Expression
- ubiquitous
- in the brain found primarily in the cerebrovascular smooth muscle cells & reactive astrocytes
