Function
- Together with KRT19, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle
- phosphorylation on Ser is enhanced during EGF stimulation & mitosis
- Ser-74 phosphorylation plays a role in keratin filament reorganization
- keratin-8 associates with keratin-18
- associates with KRT20
- when associated with KRT19, interacts with DMD
- interacts with TCHP
- interacts with HCV core protein & PNN
Structure
- O-glycosylated at multiple sites; glycans consist of single N-acetylglucosamine residues
- heterotetramer of 2 type 1 & 2 type 2 keratins
- belongs to the intermediate filament family
Compartment
Expression
- observed in muscle fibers accumulating in the costameres of myoplasm at the sarcolemma membrane in structures that contain dystrophin & spectrin
- expressed in gingival mucosa & hard palate of the oral cavity
Pathology
- defects in KRT8 are a cause of cryptogenic cirrhosis
- diagnostic pathology: recognized by CAM 5.2 antibody
