5-fluorouracil/oxaliplatin/leucovorin (FOLFOX)

Indications
- stage 3 node positive colorectal carcinoma
Dosage
- 12 cycles, every 2 weeks
- day 1:
- oxaliplatin 85 mg/m2 IV infusion in 250-500 mL D5W
- leucovorin 200 mg/m2 IV infusion in D5W over 2 hours at the same time (separate bags)
- follow with 5-fluorouracil 400 mg/m2 IV bolus over 2-4 minutes, then 600 mg/m2 IV infusion in 500 mL D5W as a 22-hour continuous infusion
- day 2:
- no oxaliplatin
- leucovorin & 5-fluorouracil as in day 1
- day 1:
Adverse effects
- 5-fluorouracil induced hyperpigmentation of palms[2] (image)
