Function
- membrane-anchored forms may play a role in cellular adhesion
- megakaryocyte-potentiating factor (MPF) potentiates megakaryocyte colony formation in vitro
- proteolytically cleaved by a furin-like convertase to generate megakaryocyte-potentiating factor (MPF), & the cleaved form of mesothelin
- both MPF & the cleaved form of mesothelin are N-glycosylated
- interacts with MUC16
Structure
- belongs to the mesothelin family
Compartment
- cell membrane; lipid-anchor, GPI-anchor Golgi apparatus
- megakaryocyte-potentiating factor: secreted
- isoform 3: secreted
Alternative splicing
- named isoforms=4; minor form, major form
Expression
- expressed in lung
- expressed at low levels in heart, placenta & kidney
- expressed in mesothelial cells
- highly expressed in mesotheliomas, ovarian cancers, & some squamous cell carcinomas (at protein level)
Pathology
- antibodies against MSLN are detected in patients with mesothelioma & ovarian cancer
- soluble form found in the sera from patients with ovarian carcinoma
