Function
- adapter protein
- negatively regulates T-cell receptor (TCR) signaling
- inhibits T-cell antigen-receptor induced activation of nuclear factor of activated T-cells
- may act by linking signaling proteins such as ZAP70 with CBL, leading to a CBL dependent degradation of signaling proteins
- interacts with phosphorylated proteins ZAP70 & CD3Z via its SH2 domain (putative)
- interacts with phosphorylated CBL via its C-terminal domain
- may be phosphorylated
- loss of C-terminal domain partially abolishes inhibitory function, but can be partially compensated by higher level of protein expression
Structure
- contains 1 SH2 domain
- contains 1 SH3 domain
Compartment
- cytoplasm
- isoform 1: cell membrane, cytoplasmic vesicle localized to plasma membrane & intracellular vesicles, including late endosomal vesicles
- isoform 2: cytoplasm
- may be cytoplasmic, not localized to membranes
Alternative splicing
- alternative initiation
- named isoforms=4
Expression
- predominantly expressed in immune system
- highest levels in peripheral blood leukocytes
- expressed in spleen, thymus & lymph nodes
- expressed in T-cells, monocytes, & at low level in B-cells
- detected in placenta, prostate, skin, retina & colon
