Introduction
Homologs of hedgehog, a secreted protein involved in establishing cell fate at several points in Drosophila development.
Function
- role in cell cycle progression
- hedgehog targets include wnt family proteins (Drosophila wingless) & TGF-beta family proteins (Drosophila decapentaplegic)[1]
- hedgehog signalling promotes transcription of cyclin E & cyclin D during Drosophila development[2]
- cilia may play a role in hedgehog signalling[3]
Pathology
- disruption of hedgehog signalling is associated with congenital anomalies of the CNS & other organ systems
- activating mutations can result in some skin cancers, including basal cell carcinoma
- activation of the hedgehog signalling pathway has been associated with tumorigenesis[4]
- tumor cells may induce hedgehog signalling in noncancerous stromal cells that surround the tumor[4]
Pharmacology
- glasdegib (Daurismo) an inhibitor of hedgehog signaling is used to treat acute myeloid leukemia
