Function

• role in base excision repair of DNA damaged by oxidation or by mutagenic agents
• acts as DNA glycosylase that recognizes & removes damaged DNA bases
  • leaves an abasic site
  • preference for oxidized pyrimidines, i.e. thymine glycol, formamidopyrimidine, 5-hydroxyuracil
  • marginal activity towards 8-oxoguanine
• AP (apurinic/apyrimidinic) lyase activity & introduces nicks in DNA
• cleaves DNA backbone by beta-delta elimination to generate a single-strand break at the site of the removed base with both 3'- & 5'-phosphates
• DNA glycosylase/lyase activity towards mismatched uracil & thymine, in particular in U:C & T:C mismatches
