Function
- role in base excision repair of DNA damaged by oxidation or by mutagenic agents
- acts as DNA glycosylase that recognizes & removes damaged DNA bases
- leaves an abasic site
- preference for oxidized pyrimidines, i.e. thymine glycol, formamidopyrimidine, 5-hydroxyuracil
- marginal activity towards 8-oxoguanine
- AP (apurinic/apyrimidinic) lyase activity & introduces nicks in DNA
- cleaves DNA backbone by beta-delta elimination to generate a single-strand break at the site of the removed base with both 3'- & 5'-phosphates
- DNA glycosylase/lyase activity towards mismatched uracil & thymine, in particular in U:C & T:C mismatches
