Function
- may regulate MET signaling cascade
- seems to decrease hepatocyte growth factor (HGF)-induced transient MAPK activation
- blocks GRB2 recruitment to MET thus suppressing the GRB2-RAS pathway
- inhibits HGF-induced proliferation of MMP1 & MMP9 expression
- oligomerization is required for interaction with MET
Compartment
- secreted (putative), cell membrane
- localized in the proximal tubules, specifically in the brush border & basolateral membrane
- isoform 3, isoform 5 & isoform 9 are membrane proteins
Alternative splicing
named isoforms=9
Expression
- highly expressed in kidney, moderately in placenta, lung, prostate, liver, & digestive system
- in the kidney, localized in the proximal tubules but not in the glomerulus or distal tubules
- detected in most of the male urogenital tract epithelia, with the exception of epididymis
Pathology
- up-regulated in patients with immunoglobulin A nephropathy (IgAN)
- no significant difference in frequencies of alleles or genotypes of tandem repeats between IgAN patients & healthy controls
