defibrotide (Defitelio)

Indications

• hepatic veno-occlusive disease

Contraindications

• anticoagulation
• fibrinolytic therapy

Dosage

• 6.25 mg/kg IV q6 hours given as a 2-hour infusion for at least 21 days, & continued until hepatic veno-occlusive disease resolution or up to 60 days of treatment

Adverse effects

• hypotension, diarrhea, vomiting, nausea, epistaxis

Mechanism of action

• pro-fibrinolytic activity
