Function

• steroid 17,20 lyase, involved in testosterone & estradiol synthesis
• conversion of pregnenolone to dehydroepiandrosterone (DHEA); 17-alpha-hydroxypregnenolone is an intermediate
• conversion of progesterone to androstenedione; 17-alpha-hydroxyprogesterone is an intermediate
• catalyzes both 17-alpha-hydroxylation & 17,20-lyase phosphorylation is necessary for 17,20-lyase, but not for 17alpha-hydroxylase activity
• involved in sexual development during fetal life & at puberty
• regulated predominantly by intracellular cAMP levels

Pathology

• deficiency associated with congenital adrenal hyperplasia type-5

Pharmacology

• inhibitor abiraterone acetate may be useful for treatment of castration-resistanct prostate cancer
