Function
- steroid 17,20 lyase, involved in testosterone & estradiol synthesis
- conversion of pregnenolone to dehydroepiandrosterone (DHEA); 17-alpha-hydroxypregnenolone is an intermediate
- conversion of progesterone to androstenedione; 17-alpha-hydroxyprogesterone is an intermediate
- catalyzes both 17-alpha-hydroxylation & 17,20-lyase phosphorylation is necessary for 17,20-lyase, but not for 17alpha-hydroxylase activity
- involved in sexual development during fetal life & at puberty
- regulated predominantly by intracellular cAMP levels
Pathology
- deficiency associated with congenital adrenal hyperplasia type-5
Pharmacology
- inhibitor abiraterone acetate may be useful for treatment of castration-resistanct prostate cancer
