Introduction
Atypical ras-like protein
Function
- regulates NF-kappa-B activity
- inhibits degradation of NFKBIB
- inhibits phosphorylation of NFKBIB (putative)
- inhibits nuclear localization of RELA NF-kappa-B subunit
- both GTP- & GDP-bound forms block phosphorylation of NFKBIB
- interacts with PEST domain in both NFKBIA & NFKBIB in vitro
- probably only interacts with NFKBIB in vivo[2]
Structure
- unlike other ras family proteins, conserved Gly & Gln in positions 13 & 65 are replaced by Leu in KBRAS1 & KBRAS2
- KBRAS1 & KBRAS2 resemble constitutively active, oncogenic forms of Ras
- belongs to the small GTPase superfamily, Ras family, KappaB-Ras subfamily
