Introduction

Atypical ras-like protein.

Function

• regulates NF-kappa-B activity
• inhibits degradation of NFKBIB
• inhibits phosphorylation of NFKBIB (putative)
• inhibits nuclear localization of RELA NF-kappa-B subunit
• both GTP- & GDP-bound forms block phosphorylation of NFKBIB
• cytoplasmic retention of p65/RELA NF-kappa-B subunit
• interacts with PEST domain in both NFKBIA & NFKBIB in vitro
• probably only interacts with NFKBIB in vivo
• in COS cells, KBRAS1 associated efficiently with IkB-beta, poorly with IkB-alpha
• forms a complex with NFKBIB & NF-kappa-B
• interacts with c-Rel

Structure

• unlike other ras family proteins, conserved Gly & Gln in positions 13 & 65 are replaced by Leu in KBRAS1 & KBRAS2
• KBRAS1 & KBRAS2 resemble constitutively active, oncogenic forms of Ras
• belongs to the small GTPase superfamily, Ras family, KappaB-Ras subfamily
