Function
- probable serine peptidase
- precise substrate specificity unclear
- does not cleave peptides after a Arg or Lys
Inhibition:
- inhibited by PMSF & prefabloc, as well as leupeptin at high concentrations
- partially inhibited by TPCK, a chymotrypsin inhibitor, & E64, a cysteine protease inhibitor
- not affected by 4-amidinophenyl-methanesulfonyl fluoride (APMSF), pepstatin or EDTA
Structure
- homodimer
- belongs to the peptidase S9A family
Compartment
Alternative splicing
named isoforms=4
Expression
- widely expressed
- highly expressed in brain, skeletal muscle, heart & kidney
Pathology
- defects in PREPL are a cause of hypotonia-cystinuria syndrome
