Function
- deubiquitinates monoubiquitinated target proteins including histone H2A & H2B
- required for proper progression through S phase & subsequent mitotic entry
- may regulate the DNA damage response (DDR) checkpoint through deubiquitination of H2A at DNA damage sites
- associates with the chromatin
- thiol-dependent hydrolysis of ester, thioester, amide, peptide & isopeptide bonds formed by the C-terminal Gly of ubiquitin
- interacts (via UBP-type domain) with H2A; the interaction is less efficient than with monoubiquitinated H2A
Structure
- both protease activity & an intact Zn+2 finger are required for H2A monodeubiquitination
- belongs to the peptidase C19 family, USP3 subfamily
- contains 1 UBP-type Zn+2 finger
