Function
- catalyzes the cleavage of the pyrrol ring of tryptophan & incorporates both atoms of a molecule of oxygen
- the enzyme is unusual in that it requires an exogenous source of superoxide
- in liver, tryptophan-2,3-dioxygenase facilitates formation of N-formylkynurenine from tryptophan
D-tryptophan + O2 <--> N-formyl-D-kynurenine
L-tryptophan + O2 <--> N-formyl-L-kynurenine
Cofactor: binds 1 heme group per subunit
Structure
belongs to the indoleamine 2,3-dioxygenase family
Expression
- found in liver & extrahepatic tissues
- induced by interferon gamma
