Clinical significance
- protein bound amylase
- due to its larger size, macroamylase may not be cleared as fast as the smaller non-protein bound amylase, thus levels may be elevated in the absence of pathology

- direct identification in serum requires ultracentrifugation or chromatography
- indirect methods:
- selective precipitation of macroamylase in a polyethylene glycol solution
- temperature sensitive amylase assay (dissociation of amylase from complexed proteins)
